Summary

Tinctures are liquid, transparent, colored medicinal preparations made by extracting plant or animal substances with alcohol, ether, wine, acetone, or mixtures of alcohol with ether or water. Unlike extracts, tinctures are prepared at room temperature without heating or evaporation.

TINCTURES, tincurae (from Latin tingere - to stain), a liquid, transparent, colored medicinal form prepared by extracting plant or animal preparations with alcohol, ether, wine, acetone, or mixtures of alcohol with ether or alcohol with water. The difference between tinctures and extracts consists in that the extraction process in preparing tinctures occurs at room temperature and tinctures are not subjected to either heating or evaporation. The diversity of extracting liquids used in preparing tinctures is due to the desire to extract all therapeutically active constituents from the plant or animal preparation, obtain them in a dissolved state, and as much as possible separate unnecessary, ballast substances. Ethyl alcohol well extracts most alkaloids and glycosides, camphor, tannins, many resins, essential oils, organic acids, partially fatty oils; poorly - proteins, carbohydrates; almost完全不 extracts most inorganic salts. Ethyl ether perfectly extracts iodine, bromine, most resins, fatty and essential oils, fats, wax, and many alkaloids. Acetone very well extracts cantharidin. Water extracts inorganic and organic salts soluble in it, acids, alkalis, alkaloid salts, carbohydrates, mucilages, etc.; poorly - fatty and essential oils, resins, free alkaloids, some organic acids, etc. Wines used for preparing tinctures - sherry, malaga, marsala, white and red wines are now completely not indicated in the preparation of medicinal forms, since wines always differ in the extreme diversity of their constituents and tinctures prepared on wine turn out to be of very different composition. The German pharmacopoeia nevertheless permits the preparation of one official wine tincture (on sherry) - Tinctura Rhei vinosa. By varying the selection of extracting liquids, the concentration and temperature at which the preparation is carried out, tinctures can be prepared in the most rational way. Prepared tinctures containing alcohol or ether in their composition possess antiseptic properties and usually do not spoil if stored in a dark place in well-stoppered bottles at ordinary temperature (Ph. VII). Corresponding to the liquid used for maceration, the tincture receives its defining name: spirituosa, aetherea, vinosa, aquosa, etc. Since most tinctures are prepared on alcohol, in prescriptions the word "Tinctura" often omits the definition "spirituosa" as being self-evident. If tinctures are prepared from one substance, simple (simplex) tinctures are obtained, and when extracting from several substances - compound (composita) tinctures. During the development of alchemy, very thick and very dark tinctures were called elixirs; solutions of extracts and other medicinal substances in tinctures were also called elixirs. The name "elixirs" has been preserved in pharmacy to the present time, e.g. Elixir viscerale Hoffmanni, Elixir amarum, etc. Somewhat less thick and colored preparations bore the name essences, quint-essences, and balms. Properly speaking, only rather lightly colored preparations were called tinctures. Even earlier, at the time of the outstanding Arab physician Avicenna (980-1076), tinctures were understood as the change and transformation (mutatio) of some body in water. Such understanding does not correspond at all to our present concept of a tincture as a medicinal form. In pharmacy, the name tinctures has survived to the present time for some medicines, although in essence these medicines represent other types of medicinal forms. Such are: Tinctura Jodi; it is now correctly called according to the indication of the international convention of 1925 the alcoholic solution of iodine, Solutio Jodi spirituosa (it should be added that it is also used for external application, m. e. t. h. 323

324 centrata); Tinctura Ferri chlorati aether-хорошо известные Бестужева капли; ФУП правильно называет это лекарство спиртно-эфирным раствором хлорного железа-Spiritus aethereus ferratus; Tinctura Ferri acetici aetherea, известная под именем Кларотовых капель, представляет собой спиртно-эфирный раствор уксусножелезной соли-Solutio Ferri acetici spirituoso-aetherea, и т. д. Современные фармакопеи довольно согласно устанавливают требования, к-рые должны быть соблюдаемы при приготовлении настоек. Советская Ph. VII указывает, что тинктуры приготавливаются или настаиванием (мацерационный способ) или извлечением в перколяторе. Вещества, служащие для извлечения из них действующих начал, должны быть мелко изрезаны или же превращены в порошок. Извлечение по мацерационному способу производится в тщательно закупоренной посуде в большинстве случаев в течение 7 суток (по германской фармакопее -10 суток), при t° 15 -20° и частом взбалтывании, после этого жидкость сливают, остаток выжимают под прессом и, смешав обе порции настойки, отстаивают ee до просветления и фильтруют. Потеря спирта (15 - 20%) или эфира, задержанных в извлекаемом веществе, не должна быть пополняема новым количеством соответственной жидкости, чтобы т. o. не уменьшить концентрации растворенных в тинктуре действующих начал. Для приготовления настойки из несильно действующих или неядовитых веществ берут одну часть вещества на пять частей извлекающей жидкости. Если же тинктуры изготовляются из сильнодействующих или ядовитых веществ, то для извлечения берут столько жидкости, чтобы получилось 10 частей настойки из одной части вещества. Большинство H., содержащих сильнодействующие или ядовитые вещества, приготовляется извлечением в перколяторе, a другие по мацерационному способу в широко-горлых склянках, банках или же глиняных горшках c хорошей глазурью. Если H. получается c большим содержанием действующих начал, чем установлено фармакопеей, то H. должна быть разбавлена извлекающей жидкостью до указанного фармакопеей стандарта; H. же c меньшим содержанием действующих начал не должны быть применяемы. Настойки должны быть прозрачны, иметь надлежащий цвет, запах и вкус тех веществ, из которых они приготовлены. При хранении многие настойки становятся более темными. Приготовление настоек ведется обыкновенно при помощи 70°-ного спирта, который легче проникает в растительные и животные клетки и поэтому полнее извлекает действующие начала, чем 90-95°-ный спирт, легко осаждающий белки клеток и тем создающий препятствие для надлежащего извлечения. Однако для некоторых тинктур, содержащих смолистые вещества и эфирные масла, рациональнее употреблять 90°-ный спирт; Tinctura Cannabis indicae, T. Capsici, Tinctura Castorei, T. Convallariae majalis, T. Myrrhae и нек-рые другие H. готовятся поэтому на 90°-ном спирте. ФУП отличает приготовление настойки из майского ландыша еще тем, что предписывает готовить H. из свежесобранного ландыша, настаивая c 90°-ным спиртом в продолжение 14 суток.-- Для проверки доброкачественности H. определяют в них количество действующих начал или производят физиол. оценку силы их действия. Когда эти данные не могут быть точными (что особенно относится к несильно действующим H.), то ограничиваются определением плотного остатка, полученного выпариванием при 100-105°, удельного веса настойки и крепости спирта в H., a также органолептическими свойствами (цвет, вкус, запах). Ph. VII указывает стандартные признаки каждой из официнальных тинктур и кроме того указывает, что в H. не должно быть солей тяжелых металлов. Ph. VII содержит 36 настоек; прилагаемый список H. дает ясное представление, что рассматриваемая лекарственная форма готовится из значительного количества различных ингредиентов. Томе (Hermann Thorns) в своем новейшем (1929) руководстве по фармации перечисляет около 190 номеров различных тинктур, из которых 41 H. официнальны по германской Ph. VI; но общее количество H., изготовляемых из разнообразных лекарственных веществ, очень велико и едва ли может быть подсчитано. Можно сказать, что почти из всех растительных препаратов приготавливали или приготовляют ту или иную настойку для мед. употребления. Такое широкое использование H. как лекарственной формы объясняется следующими обстоятельствами: в H. мы имеем сконцентрированными действующие составные части растительного или животного материала, так что данная форма выгодна прежде всего c экономической стороны; полученные настойки мало изменяются и долго могут храниться без порчи действующих веществ, при хранении занимают сравнительно небодышие пространства и при перевозках достаточно портативны; H. дают возможность использовать растительные и животные препараты в момент заготовки лекарствен, материалов, когда из них получается больший выход действующих веществ. Значение H. в фармакодинамическом отношении и в терапии тоже очень велико благодаря тому, что лекарствен, вещества находятся в H. в растворенном состоянии, т. e. в готовом ко всасыванию и реакции виде; кроме того спирт или эфир, на к-рых изготовлена та или иная H., значительно ускоряют всасывание и действие лекарственных составных частей взятой H. как при внутреннем применении, так и при наружном, употреблении H., способствуя в последнем случае более глубокому и энергичному местному воздействию на слизистые, кожу и раневые поверхности. Подкожно настойки обычно не применяют из-за их местно раздражающих и вызывающих значительную боль свойств. Внутрь H. дают до 15-20 капель на прием; следует избегать давать большие количества H. внутрь, чтобы не ввести c лекарством значительных количеств спирта или эфира, далеко небезразличных для организма. B детской практике по той же причине следует совсем исключить прописывание тинктур. He рекомендуется смешивать H. как между собой, так и c (водными) микстурами, т. к. очень часто действующие начала H. выпадают в виде осадка или мути и т. o. утрачивается значение тинктуры. Официнальные настойки по Ph. VII. Tinctura Absinthii, полынная H.: 2 ч. крупно измельченной травы полыни на 10 ч. 70°-ного спирта. Уд. в. 0,895-0,9045; сухого остатка 2,3-2,8%; спирта 66-68% по объему. Внутрь на прием 10 капель 2- 3 раза в день. Возбуждающее апетит, улучшающее пищеварение. Tinctura Aconiti, H. аконита: 10 ч. мелкого порошка клубней аконита и q. s. 70°-ного спирта. Алкалоидов 0,05%; спирта 66 - 68,5% по объему. Максимальная доза внутрь на прием 0,5, на сутки-1,5. Противоневральгическое-для местного обезболивания снаружи. Tinctura Aloes, H. сабура: 1 часть крупно измельченного сабура на 5 частей 70°-ного спирта. Уд. в. 0,947-0,950; сухого остатка 12,5-18%; спирта 58-63% по объему. Ha прием внутрь 5-30 капель. Раздражающее желудок и кишки, усиливающее отделение пищеварительных соков. TincturaAloes composita, сложная настойка сабура: 9 ч. сабура толченого, 1 ч. крупно истолченного корня горечавки, 1 ч. крупно истолченного корневища ревеня, 1 ч. шафрана, 1 ч. крупно истолченного корневища зедоарии на 200 частей 70°-ного спирта. Внутрь по 1/г-1 чайной ложке несколько раз в день. Возбуждающее апетит, улучшающее пищеварение. Tinctura amara, горькая H.: 2 ч. крупно истолченных незрелых плодов померанца, 2 ч. мелко изрезанной травы золототысячника, 2 ч. крупно измельченного корня горечавки, 1 ч. крупно истолченного корневища зедоарии на 35 ч. 70°-ного спирта. Удельный вес 0,905-0,907; сухого остатка 4-6%; спирта 65-68% по объему. Ha прием внутрь 20 - 50 капель несколько раз в день. Возбуждающее апетит, улучшающее пищеварение. Tinctura Arnicae, H. арники: 1 ч. изрезанных цветов арники на 10 ч. 70°-ного спирта. Уд. в. 0,898-0,905; сухого остатка 1,5-2%; спирта 66-68% по объему. Внутрь на прием 10-30 капель 2-3 раза в сутки, снаружи в чистом виде или в разведении c водой в форме компресов или примочек при ушибах, ранениях, невральгиях. Tinctura Aurantii, H померанцевой корки: 2 ч. крупно измельченной корки, очищенной от белой мякоти, на 10 ч. 70°-ного спирта. Уд. в. 0,910-0,917; сухого остатка 4-5%; спирта 65-67% по объему; внутрь на прием 1,0-4,0 несколько раз в день. Исправляющее вкус для микстур. Tinctura Belladonnae, H. ночной красавки или сонной одури: 10 ч. порошка листьев белладонны и q. s. 70°-ного спирта. Алкалоидов 0,035% ; спирта 67 - 69% по объему. Внутрь максимальная доза на прием 0,5, в сутки-1,5; снаружи в клизмах. Болеутоляющее, противоспазматическое. Tinctura Cannabis indicae, H. индийской конопли: 10 ч. крупно измельченной травы индийской конопли и q. s, 90°-ного спирта. Уд. в. 0,839-0,843; спирта-87-88% по объему. Внутрь максимальная доза на прием 1,25, в сутки-3,75. Болеутоляющее при метрорагиях и менорагиях. Tinctura Cantharidis, H. шпанских мух: 10 ч. крупно истолченных шпанских мух и q. s. 70°-ного спирта. Удельный вес 0,893-0,904; сухого остатка 1,5-2,2%; спирта 67-69% по объему. Внутрь максимальная доза на прием 0,2 (3 капли), в сутки 0,6; излишний и опасный препарат; в форме втираний, линиментов, мазей, помад как раздражающее кожу - для усиления роста волос.

' Tinctura Capsici, H. of red pepper: 1 part finely chopped red pepper with seeds to 10 parts of 90° alcohol. Specific gravity 0.837 - 0.843; dry residue 1.42-1.58%; alcohol 86-88% by volume. Internally, per dose 0.5-1.5 several times a day to stimulate stomach activity; externally, in the form of mouth and throat gargles 1.0 - 10.0 : 100.0 and for rubbing as an irritant. Tinctura Castorei, H. of castoreum: 1 part coarsely crushed and dried castoreum to 10 parts of 90° alcohol. Specific gravity 0.845-0.849; dry residue 3.45-4.65%; alcohol 86 - 88% by volume. Internally, per dose 1.0 - 3.0 as a stimulant of nervous activity. Tinctura Chinae, cinchona H.: 10 parts coarsely crushed cinchona bark to 100 parts of 70° alcohol. Alkaloids 0.74%; dry residue 4-6%; alcohol 64-66% by volume. Internally 1.0-4.0 two to three times a day. Appetite stimulant. Tinctura Chinae composita, compound cinchona H.: 6 parts coarsely crushed cinchona bark, 1 part coarsely crushed cinnamon, 2 parts coarsely crushed orange peel, 2 parts coarsely crushed gentian root to 50 parts of 70° alcohol. Alkaloids 0.37%; dry residue 6-8%; alcohol 63 - 65% by volume. Internally 1.0 - 5.0 two to three times a day. Appetite stimulant, improves nutrition. Tinctura Cinnamomi, H. of cinnamon: 2 parts coarsely crushed cinnamon to 10 parts of 70° alcohol. Specific gravity 0.895-0.905; dry residue 1.2-2%; alcohol 65-68% by volume. Internally 1.0-3.0 several times a day. Stimulates digestion; externally as an additive to mouthwashes, dental tinctures and dental elixirs. Tinctura Convallariae majalis, H. of lily of the valley: 100 parts crushed fresh lily of the valley herb to 120 parts of 90° alcohol. Specific gravity 0.925-0.927; dry residue 2.2-3%; alcohol 56-59% by volume. Per dose maximum 1.25, daily maximum 3.75. Used in cardiac nervous diseases.

\ Tinctura Digitalis, H. of foxglove: 10 parts coarsely crushed foxglove leaves and q.s. 70° alcohol. Tincture strength-4 units of action in 1 g of tincture; specific gravity 0.9025-0.907; dry residue 2.5 - 3.2%; alcohol 66-69% by volume. Internally, per dose maximum 1.0/daily maximum 3.0. Cardiac remedy.

11 Tinctura Ferri pomati, H. of iron malate: 1 part of iron malate extract to 9 parts of cinnamon alcoholic water. Iron 0.4-0.5% in the form of salts; specific gravity 1.021-1.030; dry residue 6-9.2%. Internally 1.0-3.0 several times a day; for anemia. Tinctura Gallarum, H. of galls: 1 part coarsely crushed galls to 5 parts of 70° alcohol. Specific gravity 0.945-0.958; dry residue 12.5-14.6%; alcohol 61-64% by volume. Internally 0.75-2.0 several times a day as an astringent; externally (also as an astringent) for mouth rinsing, gum application, in enemas. Tinctura Gentianae, H. of gentian: 2 parts coarsely crushed gentian root to 10 parts of 70° alcohol. Specific gravity 0.916-0.920; dry residue 6-6.5%; alcohol 63-67% by volume. Internally 1.0-3.0 several times a day. Appetite stimulant, improves digestion. Tinctura Jodi, iodine tincture: 1 part of sublimed iodine to 9 parts of 95° alcohol. Iodine 9.5-10%; specific gravity 0.887-0.891. Internally, per dose maximum 0.2, daily maximum 0.6; externally in cases where iodine is indicated. Tinctura Ipecacuanhae, ipecac tincture: 10 parts of fine powder of ipecac root and q.s. 70° alcohol. Alkaloids 0.2%; alcohol 67 - 69% by volume. Internally, per dose maximum 2.0, daily maximum 4.0, as an expectorant and emetic; disinfectant for amebic dysentery. Tinctura Menthae piperitae, mint H.: 1 part chopped leaves of peppermint, 1 part peppermint oil to 20 parts of 90° alcohol. Internally 5-10 drops several times a day; corrects taste for mixtures, cooling and flavoring for gargles. Tinctura Myrrhae, H. of myrrh: 1 part myrrh powder to 5 parts of 90° alcohol. Specific gravity 0.843-0.856; dry residue 4.24-5.26%; alcohol 84-87% by volume. Externally for mouth rinses.

, Tinctura Opii benzoica, benzoic-opium H.: 1 part anise oil, 2 parts camphor, 4 parts benzoic acid, 10 parts opium tincture on 183 parts of 70° alcohol (morphine 0.05%). Specific gravity 0.891-0.901; alcohol 66-67% by volume. Internally, per dose 1.0-4.0 several times a day; calming for cough.

> Tinctura Opii crocata, saffron-opium tincture: 15 parts opium in powder, 1 part coarsely crushed cinnamon, 1 part coarsely crushed cloves, 5 parts coarsely crushed saffron, 70 parts water to 70 parts of 70° alcohol (morphine 1%). Internally, maximum dose per administration 1.0, daily 3.0. Analgesic, sedative, antispasmodic, antidiarrheal. Temporarily discontinued from production. Tinctura Opii simplex, opium tincture: 4 parts opium powder, 19 parts 70° alcohol, and 19 parts water (morphine 1%). Internally, maximum dose per administration 1.0, daily 3.0; analgesic, sedative, antispasmodic, antidiarrheal. Tinctura Polygoni hydropiperis, tincture of water pepper: 2 parts coarsely powdered water pepper herb to 10 parts 70° alcohol. Sp. gr. 0.897-0.902; dry residue 2-3%; alcohol 66-68% by volume. Internally 1.0-2.0 several times a day. Hemostatic for uterine, hemorrhoidal bleeding, etc. Tinctura Ratanhiae, tincture of ratanhia: 2 parts coarsely powdered ratanhia root to 10 parts 70° alcohol. Sp. gr. 0.910-0.920; dry residue 4.5-6%; alcohol 64-66% by volume. Internally 1.0-1.5 several times a day as an astringent, and externally for mouth rinsing and gum application. Tinctura Rhei amara spirituosa, bitter alcoholic tincture of rhubarb: 8 parts coarse powder of gentian root, 32 parts coarse powder of rhubarb rhizome, 3 parts coarse powder of serpentaria rhizome to 384 parts 70° alcohol. Sp. gr. 0.900-0.908; dry residue 3-4.5%; alcohol 65-68% by volume. Internally 2.0-4.0 several times a day. Laxative, regulating stomach and intestinal activity. Tinctura Rhei aquosa, aqueous tincture of rhubarb: 10 parts cut rhubarb rhizome, 1 part borax, 1 part pure potassium carbonate, 90 parts boiling water, 9 parts 90° alcohol, 15 parts cinnamon water. Specific gravity 1.008-1.012; dry residue 4.1-4.5%. Internally 2.0-10.0 several times a day; regulating intestinal activity; 15.6-60.0 as a laxative (unreliable). Tinctura Strophanthi, tincture of strophanthus: 10 parts coarsely powdered strophanthus seeds and q.s. 70° alcohol. Valence 200-250 units of action in 1 g of tincture. Sp. gr. 0.893-0.899; dry residue 1.5-2%; alcohol 65-68% by volume. Internally, maximum dose per administration 0.5, daily 1.5; cardiac. Tinctura Strychni, tincture of nux vomica: 10 parts nux vomica seed powder and q.s. 70° alcohol. Alkaloids 0.25%; alcohol 67-69% by volume. Internally, maximum dose per administration 1.0, daily 3.0. Improves digestion, enhances stomach and intestinal movement as a tonic; also in paresis, paralysis, in narcotic poisoning, in alcoholism, in cardiac weakness. Tinctura Valerianae, valerian tincture: 1 part coarsely powdered valerian rhizome and root to 5 parts 70° alcohol. Sp. gr. 0.894-0.901; dry residue 2-3%; alcohol 65-67% by volume. Internally 1.0-3.0 several times a day; sedative for nervous disorders, stimulates cardiac activity. Tinctura Valerianae aethera, ether-valerian tincture: 1 part coarsely powdered valerian rhizome and root, 4 parts 90° alcohol and 2 parts ether. Sp. gr. 0.805-0.809; dry residue 1-1.6%; alcohol 64-65% by volume. Internally 1.0-1.5 several times a day. Sedative for nervous disorders, stimulates cardiac activity. Tinctura Veratri, tincture of white hellebore: 10 parts white hellebore rhizome powder and q.s. 70° alcohol. Sp. gr. 0.896-0.910; dry residue 1.39-2%. Internally, maximum dose per administration 1.0, daily 3.0; superfluous and dangerous preparation. To tinctures can also be attributed the following medicines indicated in the FUP. Acetum aromaticum, aromatic vinegar: 2 parts bergamot oil, 2 parts lemon, 2 parts thyme, 1 part eucalyptus, 1 part rosemary oil, 20 parts acetic ether, 350 parts 90° alcohol, 384 parts diluted acetic acid, 1728 parts water, 20 parts powdered cinnamon bark and 20 parts powdered cloves. Tincture-sp. gr. 0.985-0.988; externally-for disinfecting washes, for mouth rinsing. Guttae Botkini, Botkin's drops: 120 parts of complex quinine tincture, 120 parts of Hoffman's drops, 23 parts of diluted hydrochloric acid, 15 parts of hydrochloride quinine, 2 parts of peppermint oil. Tincture-sp. gr. 0.895-0.910; 7.5-8% dry residue. Internally 1.0-1.5 several times a day. Analgesic for stomach diseases. Guttae Inosemzowi, Inozemtsev's drops: 1 part nux vomica extract, 120 parts opium tincture, 480 parts rhubarb tincture, 20 parts castoreum tincture, 240 parts Hoffman's drops, 240 parts ether-valerian tincture, 5 parts peppermint oil. Tincture-sp. gr. 0.871-0.877; 2.5-3% dry residue; internally 0.5-1.0 several times a day. Analgesic for stomach diseases, antidiarrheal. Due to the temporary cessation of import of some types of medicinal raw materials from abroad, prescriptions containing imported raw materials have been changed by a circular of the People's Commissariat of Health, and some tinctures (orange, kola nut, etc.) are temporarily not manufactured. Iodine tincture is temporarily prepared with 5% iodine content. V. Nikolaev.
